Mechanism of production of age spots(melanin)

The skin consists of the epidermis and the dermis, the epidermis consists of four layers: the stratum corneum, the granule layer, the spine layer, and the basal layer, in order from the surface. Melanin is produced by pigment cells (melanocytes) located in the basal layer between the epidermis and the dermis and is transported to nearby epidermal cells. Age spots are formed by the accumulation of melanin in the epidermis and are caused by prolonged exposure to UV light or by disturbances in the metabolic rate (turnover) of the skin.

Using cosmetic products containing ozonized glycerin, we verified the effect on surfaced age spots and conducted effects on melanin in human clinical trials.

【Test Method】

Using cosmetic products containing ozonized glycerin and glycerin without ozonized glycerin (hereinafter referred to as a control product), two pushes in the morning and evening are applied to the half face twice a day everyday. Four or eight weeks later, we photographed facial images with VISIA-Evolution and measured melanin content using a mexameter (melanin measuring instrument).

【Test Results】

It was confirmed that melanin levels decreased on the skin surface of about 92% of those who continued to use ozonized glycerin-containing samples for 8 weeks (Fig. 2). After 8 weeks of continuous use of samples containing ozonized glycerin, it was confirmed that the melanin value decreased by about 20% on average (Fig. 3).

  • (2) Effect of reducing age spots(senile lentigos) on the facial skin

Using a sample containing ozonized glycerin, we confirmed that it is also effective for age spots(senile stains) on the skin surface that have already appeared for a long time.

Ozonized glycerin can be expected to work not only on the age spots reserves that will surface in the future, but also on “age spots” that have surfaced on the skin for a long time (Fig. 4).

  • (3) Melanin direct resolution*³ test of high performance ozonized glycerin

Continuous use of samples containing ozonized glycerin for 8 weeks has confirmed the effect of thinning age spots and blemishes in human clinical trials. Therefore, in order to elucidate the mechanism by which ozonized glycerin reduces age spots, we conducted tests to explore the possibility of “direct decomposition of melanin*³” of ozonized glycerin.

【Test Method】

A melanin solution was prepared and ozonized glycerin and five types of other skin blighting solvents were used to compare changes in melanin pigments in 1, 2, and 4 weeks.

【Test Results】

Compared to five types (vitamin C, hydroquinone, kojic acid, niacinamide, tranexamic acid), which are mainly famous as blighting active ingredients, The ozonized glycerin has the effectiveness of Melanin direct dissolution *³” was confirmed (Figs. 5 and 6). It was confirmed that the concentration (depth) of melanin by 66% was reduced in 4 weeks (Fig. 7).

The results suggest that ozonized glycerin formulations could lighten the age spots (melanin) to an advantage in 8 weeks, and that one of the mechanisms could be to directly decompose melanin already deposited in the facial skin(B).

“Ozonized glycerin”, which is extremely safe to the skin and has the possibility of direct decomposition of melanin, will become a next-generation skin care active ingredient that will attract more and more attention in the future.
